Jaumet reduces the level of glucose (sugar) in the blood. This medicine has two components: sitagliptin, a medicine that transports formed insulin to the pancreas, and metformin, a substance that helps the liver and muscles to better absorb glucose from the blood. Janumet, therefore, stabilizes blood glucose content in patients with diabetes. Janumet is prescribed to patients with type 2 diabetes. Insulin is not produced sufficiently in this form of diabetes. Also, the body does not respond adequately to insulin. Insulin is important in the exchange of sugar. In diabetes patients who do not produce enough hormone, the glucose level increases too much. This causes complaints such as thirst, tiredness, frequent urination, and poorly healing wounds. If diabetes is not treated, serious long-term complications can arise. Janumet stimulates the formation of insulin and therefore prevents the glucose content in the blood from increasing too much.

Janumet is a prescription medicine that is used to help manage blood sugar levels in patients who have type 2 diabetes. In its tablet format, Janumet contains the active ingredients Sitagliptin and Metformin, which, when combined, can help to improve insulin sensitivity and lower the production of glucose within the liver, thus regulating blood sugar control. Janumet can be prescribed as a standalone treatment or in combination with other diabetes medicines to help improve and balance blood sugar levels.
What is Janumet?
Janumet contains two different medicines, known as sitagliptin and metformin, which when used in conjunction with a healthy diet and exercise routine will control increased blood sugar in type 2 diabetes. Metformin belongs to a class of medicines called biguanides and Sitagliptin belongs to a class of medicines called DPP-4 inhibitors (dipeptidyl peptidase-4 inhibitors).

How does it work?
Sitagliptin phosphate is a highly selective, potent, and orally active inhibitor of the enzyme dipeptidyl peptidase 4 (DPP-4) for the treatment of type 2 diabetes. DPP-4 inhibitors are a class of agents that act as enhancers. of incretin. By inhibiting the enzyme DPP-4, sitagliptin increases the levels of two known active incretin hormones, glucagon-like peptide-1 (GLP-1) and glucose-dependent insulinotropic polypeptide (GIP). Incretins are part of an endogenous system involved in the physiological regulation of glucose homeostasis. When blood glucose concentrations are normal or elevated, GLP-1 and GIP increase insulin synthesis and release from pancreatic beta cells. GLP-1 also reduces glucagon secretion from pancreatic alpha cells, leading to reduced hepatic glucose production. When blood glucose levels are low, insulin release is not enhanced and glucagon secretion is not suppressed. Sitagliptin is a highly selective and potent inhibitor of the enzyme DPP-4 and, at therapeutic concentrations, does not inhibit the closely related enzymes DPP-8 or DPP-9. Sitagliptin differs in chemical structure and pharmacological action from GLP-1 analogues, insulin, sulfonylureas or meglitinides, biguanides, peroxisome proliferator-activated gamma receptor (PPAR?) Agonists, alpha inhibitors -Glucosidase and amylin analogues.

How do I use it and its dosage?

For the recommended Janumet Sitagliptin dosage, take 2 tablets a day orally, with food to reduce the likelihood of stomach discomfort
Your doctor may need to increase the dose to control your blood sugar levels.
If you have reduced kidney function, your doctor may prescribe a lower dose.
You should continue the diet recommended by your doctor during treatment with this medicine and take care that the carbohydrate intake is distributed equally throughout the day.
Treatment with this medication alone is unlikely to cause an abnormal drop in blood sugar (hypoglycemia). A drop in blood sugar can occur when this medicine is taken together with a medicine that contains sulfonylurea or insulin, so your doctor will probably find it necessary to reduce the dose of your sulfonylurea or insulin.
Side effects & precautions
Like all medicines, these Diabetes Treatment medicines can cause side effects, although not everybody gets them.
STOP taking Janumet and see a doctor immediately if you notice any of the following serious side effects:
Persistent and severe pain in the abdomen (stomach area) that can reach the back with or without nausea and vomiting, as these may be signs of inflammation of the pancreas (pancreatitis).
Janumet can cause a very rare (may affect up to 1 in 10,000 people) but a very serious side effect, called lactic acidosis. If this happens to you, you should stop taking Janumet and contact a doctor or the nearest hospital immediately, as lactic acidosis can lead to a coma.
If you have a severe allergic reaction (frequency not known), including a rash, hives, skin blisters / peeling skin, and swelling of the face, lips, tongue, and throat that can cause difficulty breathing or swallowing, stop take this medication and consult your doctor immediately. Your doctor will prescribe a medicine to treat the allergic reaction and will change the medicine to treat diabetes.
Some patients taking metformin have experienced the following side effects after starting sitagliptin treatment:
- Common (may affect up to 1 in 10 people): low blood sugar, nausea, gas, vomiting
- Uncommon (may affect up to 1 in 100 people): stomach pain, diarrhea, constipation, drowsiness
Some patients have experienced diarrhea, nausea, gas, constipation, stomach pain or vomiting when starting treatment with the combination of sitagliptin and metformin (frequency classified as frequent).
Some patients have experienced the following side effects when taking this medicine together with a sulfonylurea such as glimepiride:
- Very common (may affect more than 1 in 10 people): low blood sugar levels
- Common: constipation
Some patients had the following side effects while taking this medicine in combination with pioglitazone:
- Common: swelling of hands or legs
