How does it work?

Saxagliptin is a highly potent, selective, reversible, and competitive DPP4 (Ki: 1.3 nM) inhibitor. Administration of saxagliptin in patients with type 2 diabetes inhibited the enzymatic activity of DPP4 for a period of 24 hours. Following oral glucose overload, this DPP4 inhibition caused a 2 to 3-fold increase in circulating levels of active incretin hormones, including glucagon-like peptide 1 (GLP-1) and glucose-dependent insulinotropic polypeptide (GIP). , a decrease in glucagon concentrations and an increase in the response capacity of beta glucose-dependent cells, thereby increasing insulin and C-peptide concentrations. The increase in insulin produced by the beta cells of the pancreas and the decrease Glucagon levels produced by pancreatic alpha cells was associated with lower fasting glucose concentrations and reduced fluctuation in glucose levels after oral glucose overload or after a meal. Saxagliptin improves glycemic control by reducing fasting and postprandial glucose concentrations in patients with type 2 diabetes.

What are the benefits of taking it?

Onglyza uses include being indicated to treat adult patients 18 years of age and older with "type 2 diabetes" if the disease cannot be adequately controlled with oral anti-diabetic medication, diet, and exercise. Onglyza is used alone or together with insulin or other antidiabetic medicines. It is important that you follow the diet and exercise advice given by your doctor or healthcare staff.

How do I use it and its dosage?

Always use this medicine exactly as your doctor has told you. If in doubt, consult your doctor or pharmacist again.

The recommended dose is one 5 mg tablet once a day.

If you have decreased kidney function, your doctor may prescribe a lower dose. This is one 2.5 mg tablet once a day. For this dose, a different strength tablet is available.

The doctor may prescribe Onglyza alone or together with insulin or other anti-diabetic medications. If applicable, remember to take these other medications as directed by your doctor to achieve the best health results.

How to take Onglyza

The tablets should not be divided or cut. Swallow the tablet whole with a little water. You can take the tablet with or without food. The tablet can be taken at any time of the day, however, try to take the tablet at the same time each day. This will help you remember to take it.

If you take more Onglyza than you should

If you take more tablets than you should, see a doctor immediately.

If you forget to take Onglyza

If you forget to take a dose of Onglyza, take it as soon as you remember. However, if your next dose is not long, do not take the missed dose.

Do not take a double dose to make up for a forgotten dose. Never take two doses on the same day.

Side effects & precautions

Like all medicines, this medicine can cause side effects, although not everybody gets them.

Some symptoms need immediate medical attention:

You should stop taking Onglyza and see your doctor immediately if you experience the following symptoms of low blood sugar: tremor, sweating, anxiety, blurred vision, tickling of the lips, paleness, mood changes, disorientation or confusion (hypoglycemia); very frequently observed (may affect more than 1 in 10 patients).

Symptoms of a severe allergic reaction (rarely seen, may affect up to 1 in 1,000 patients) may include:

  • Acne
  • Raised red welts on the skin (hives)
  • Swelling of the face, lips, tongue, and throat that can cause difficulty breathing or swallowing.
  • If you experience any of these symptoms, stop taking Onglyza, and contact your doctor or nurse immediately. Your doctor may prescribe a medicine to treat your allergic reaction and a different medicine for your diabetes.

You should stop taking Onglyza and contact your doctor immediately if you notice any of the following serious side effects:

severe and persistent pain in the abdomen (in the stomach area) that can reach the back, as well as nausea and vomiting, which could be a sign of having an inflamed pancreas (pancreatitis).

You should contact your doctor if you experience the following side effect:

  • Severe joint pain

Some patients have had the following side effects while taking Onglyza and metformin:

  • Common Saxagliptin Side Effects (may affect 1 in 100 patients): chest or lung infection, urinary tract infection, inflammation of the stomach or intestine normally caused by an infection (gastroenteritis), sinus infection with pain and pressure behind the cheekbones and eyes (sinusitis), inflammation of the nose or throat (nasopharyngitis) (signs of this may include a cold or sore throat), headache, muscle pain (myalgia), vomiting, inflammation of the stomach (gastritis), stomach pain and indigestion (dyspepsia).
  • Uncommon Saxagliptin Side Effects (may affect 1 in 1,000 patients): joint pain (arthralgia) and difficulty having or maintaining an erection (erectile dysfunction).

Some patients have had the following side effects while taking Onglyza and a sulfonylurea:

  • Very common: low blood sugar levels (hypoglycemia).
  • Common: chest or lung infection, urinary tract infection, inflammation of the stomach or intestine, usually caused by an infection (gastroenteritis), sinus infection with pain and pressure sensation behind the cheekbones and eyes ( sinusitis), headache, stomach pain, and vomiting.
  • Uncommon: tiredness, abnormal levels of lipids (fatty acids) (dyslipidemia, hypertriglyceridemia).

Some patients have had the following side effects while taking Onglyza and a thiazolidinedione:

  • Common: chest or lung infection, urinary tract infection, inflammation of the stomach or intestine, usually caused by an infection (gastroenteritis), sinus infection with pain and pressure sensation behind the cheekbones and eyes ( sinusitis), headache, vomiting, stomach pain, and swelling of the hands, ankles, or feet (peripheral edema).

Some patients have had the following side effects while taking Onglyza and metformin and a sulfonylurea:

  • Common: dizziness, tiredness, stomach pain, and flatulence.

Some patients have had the following additional side effects while taking Onglyza alone:

  • Common: dizziness, diarrhea, and stomach pain.

Some patients have had the following side effects while taking Onglyza alone or in combination:

  • Not known (frequency cannot be estimated from the available data): constipation, blisters on the skin (bullous pemphigoid).

Some patients have had a small reduction in the number of a type of leukocytes (lymphocytes) detected by blood tests while taking Onglyza alone or in combination.
