Constella (Linaclotide) is a gastrointestinal medication that is indicated for the symptomatic treatment of irritable bowel syndrome (IBS) syndrome with moderate to severe constipation in adults. IBS is a chronic disorder of intestinal function characterized by abdominal pain or discomfort, accompanied by swelling and changes in the gastrointestinal tract. How does it work?

Both linaclotide and its active metabolite bind to the guanylate cyclase-C (GC-C) receptor, on the luminal surface of the intestinal epithelium. The action of linaclotide on the GC-C receptor has been shown to reduce visceral pain and intensify GI transit in animal models and increase colonic transit in humans. GC-C activation results in increased concentrations of cyclic guanosine monophosphate, both extracellularly and intracellularly.
How do I use it and its dosage?
Always use this IBS medication exactly as your doctor has told you. If in doubt, consult your doctor or pharmacist again. The recommended dose is one capsule (that is, 290 micrograms of linaclotide) by mouth once a day. The capsule should be taken at least 30 minutes before a meal. The doctor should periodically assess the need to continue treatment. The effectiveness of linaclotide was demonstrated in double-blind studies of up to six months and placebo-controlled. If patients have not experienced an improvement in symptoms after four weeks of treatment, it is recommended that they undergo a further examination and that reconsider the benefits and risks of continuing to administer the medication.
Special populations
Patients with kidney or liver failure
No dosage adjustment is necessary for patients with hepatic or renal impairment. Elderly patients
In elderly patients, although no dose adjustment is necessary, treatment should closely watched and checked periodically.
Pediatric population
The safety and efficacy of Constella in children aged 0-18 years have not yet been established. Therefore, this medicine should not be used in children or adolescents.}
